Preparation:
Agarose Dish
- Prepare 1% or 1,5% (w/v) agarose dishes.
- Either 1 or 1,5gr agarose in 100ml eggwater (0,6g instant Ocean in 10L water)
- Use a blue capped bottle (at least 250ml volume to allow for extra space during boiling)
- Boil in a microwave (make sure to remove the cap of the bottle while boiling!!)
- Boil for 30-60s and check the turbidity of the agarose solution (repeat until the solution is clear)
- Pour app 25ml hot agarose solution per pertridish.
- Allow for the agarose to solidify (app. 10-20min)
- Use immediately, or store at 4°C in inverted position.
- When using dishes that have been stored for a long time make sure to add a small volume of eggwater to the dishes before starting injection (this will prevent the fish from drying out to fast).
Needles:
Make sure that the capillaries have been calibrated on the filament you are using, when you switch either filament or capillary you should determine the ramp value of the capillaries (switch on needle puller, select program, clear, select no, select ramp value, place a capillary and press pull)
- Before making needles check the structural integrity of the filament (small glowing part) of the needle puller.
- When the filament looks different (not square anymore, holes, molten etc) contact someone who can change the filament, or change the filament yourself.
- Turn on the needle puller, select a program (#99,#4 or make a new specialized program to your liking, program #99, Heat=ramp+15, pull=95, vel=60, time=90)
- Place the a capillary (0,78OD) in the holder on one side of the filament guard.
- Move the capillary towards the filament, making sure to stop before entering the filament guard.
- Tighten the screw on the filament arm.
- Move the arm towards the center. Depress the clamp to enable the arm to move.
- When the arm is fully towards the center unscrew the filament arm slightly allowing the capillary to move.
- Move the opposite arm towards the center and move the capillary through the filament guard.
- Center the capillary.
- Tighten the screws at both ends of the filament.
- Press the pull button and wait for the needle puller to finish.
- Open the needle clamps on both sides and remove the needles in an upward fashion.
- Place the needles in a designated Petri dish (containing either clay or tape to stick the needles to)
